DusQ: New dark quencher series from Lumiprobe
Dark quenchers are non-fluorescent chromophores that can absorb excitation energy from a fluorophore and dissipate it as heat. Quenchers are widely used to synthesize oligonucleotides: hydrolyzable DNA probes, «molecular beacons», and «scorpion» primers. The use of quenchers in these applications has many advantages:
- low background fluorescence and better signal-to-noise ratio;
- high dynamic range;
- possibility of multiplexing since quenchers have no secondary fluorescence;
- straightforward synthesis of DNA probes since quenchers are resistant to degradation at the deprotection stage.
We offer a new series of DusQ® fluorescence quenchers with different spectral properties and functional groups for oligonucleotide synthesis and other applications.
DusQ® 1DusQ1 has an absorption maximum of 522 nm and an effective 480–580 nm absorption range. This dark quencher is preferred for use with fluorescent dyes in the yellow-green or yellow part of the spectrum (519–556 nm), such as FAM and TET. |
DusQ® 2DusQ2 has an absorption maximum of 552 nm and an effective 560–670 nm absorption range. This dark quencher effectively quenches the fluorescence of dyes in the yellow-orange part of the spectrum (557–617 nm), such as HEX, JOE, TMR, TR, Cyanine3, and ROX. |
DusQ® 3DusQ3 has an absorption maximum of 657 nm and an effective absorption range of 620–730 nm. This dark quencher is recommended for use with red and far-red fluorescent dyes (557–617 nm), such as BDP630/650, Cyanine5, Cyanine5.5, and AF 647. |
DusQ® 21DusQ21 has an absorption maximum of 656 nm and an effective absorption range of 690–720 nm. This dark quencher can be used as an alternative to DusQ3 for coupling with red and far-red fluorescent dyes (557–617 nm), such as BDP630/650, Cyanine5, Cyanine5.5, and AF 647. |
