4× DNA/RNA Gel Loading Buffer is used to prepare nucleic acid samples for loading into an electrophoresis agarose gel .
The buffer contains Bromophenol Blue and Xylene Cyanol FF dyes and allows monitoring the mobility of the DNA/RNA fragments being analyzed in the gel. In a 1% TAE-agarose gel, the Bromophenol Blue front migrates at the level of nucleic acid fragments approximately 300-400 bp in size, while Xylene Cyanol migrates at approximately 3000-4000 bp.
The buffer also contains EDTA to inhibit metal-dependent nucleases and prevent nucleic acids degradation, and glycerol as a weighting agent to ensure uniform sample loading at the bottom of the gel well.
For use, add 4× DNA/RNA Loading Buffer to the DNA or RNA sample (1/3 of the sample volume), mix, and load into the agarose gel.
