Vacuolin-1 is a triazine-based small chemical, a specific inhibitor of Ca2+–dependent lysosomal exocytosis. The inhibitor penetrates the cell, blocking the fusion of lysosomes with the plasma membrane. Vacuolin-1 does not affect other membrane-bound organelles and endocytic or secretory traffic. The inhibitor is widely used in studies on the release of lysosomal content and the modulation of autophagy.
