Tetraacetylated N-(4-pentynoyl)galactosamine (Ac4GalNAl) is an alkyne-labeled monosaccharide designed for metabolic labeling of galactosamine-containing glycoconjugates. Due to its acetylated groups, the compound readily penetrates cells, where it is deacetylated and integrated into biosynthetic glycosylation pathways instead of natural N-acetylgalactosamine (GalNAc).
Glycoconjugates containing the incorporated alkyne can be selectively visualized or affinity enriched using Cu(I)-catalyzed (CuAAC) click reaction with fluorescent or biotinylated azides, making Ac4GalNAl a valuable tool for studying the structure and dynamics of O-galactosaminylated biomolecules.
