Azide-PEG6-hydrazide is a heterobifunctional crosslinker with a hydrazide group, alkyne-reactive azide group, and a PEG4 hydrophilic linker.

The hydrazide group is used to attach the terminal azide group to a carbonyl group of aldehydes and ketones. Carbonyl groups are encountered in proteins subjected to oxidative stress. Many small natural molecules are also carbonyl compounds. Most saccharides and RNAs contain 1,2-diol fragments, which can be oxidized with periodate to aldehydes, which in turn react with hydrazides.

The azide group can undergo click chemistry reactions with alkyne, BCN, or DBCO to form a stable triazole linkage.
